The research aimed to investigate the changes of some essential bioactive compounds in lotus leaves (Nelumbo nucifera Gaernt) during the extraction process. Total phenolic content (TPC) and total flavonoid content (TFC) were measured by the Folin - Ciocalteu method and the aluminum chloride colorimetric assay while the antioxidant capacity was determined by DPPH free radical scavenging technique. The basic factors were evaluated as type of solvents (hot water, propanol and ethanol at various concentrations 50-100% v/v), the ratio of material and solvent (1:10, 1:20, 1:30, 1:40) and various extraction temperature (40, 50, 60, 70, 80oC). The results reported that highest bioactive activity were obtained when being extracted in ethanol 70% v/v with the ratio 1:30 (lotus leaves: solvent) at the optimal temperature 70oC, particularly the amount of TPC reached 5075.95 ± 51.05 mg gallic acid equivalent/100 gram of dry material and TFC (3125.20±21.60 mg quercetine equivalents/100 gram of dry material) and antioxidant capacity (62.16±0.37 %).
