One study to find the appropriate nutrients to culture Bacillus isolates from the local area to serve aquaculture was done. The study consists of two experiments. Experiment 1 was carried out with 3 types of nutrient medium including sugar, TSB (Tryptone Soya Broth) and Glucose. Experiment 2 to determine the optimal nutrient medium (from the experiment 1) to storage bacteria at different temperature: room conditions (28°C) and fridge conditions (10°C). Each experiment was performed with three strains of Bacillus: Bacillus sp (B2), Bacillus amiloliquefaciens (B41) and Bacillus subtilis (B67). The experiment was repeated 3 times arranged completely random in vitro.
The results from Experiment 1 showed the density of Bacillus get highest in treatments TSB (3.4 ì 108 CFU/ml), and lowest in sugar and glucose treatment (1.6 ì 108 CFU/ml), and molasses (1.7 ì 108 CFU/ ml) respectively. However, differences between the treatments were not significant (p>0.05). Therefore sugar can be as optimal liquid nutrient medium for Bacillus; because its is inexpensive, easy to implement for farmers. Results from Experiment 2 also showed that although in the cooler conditions (10°C ) can preserve biomass Bacillus better, but it was not significant difference (P>0.05) with the storage temperature (28°C).
