Adhering microflora profile is essential and used for the development and design of the cleaning and sanitizing procedures, which is a part of the HACCP system. The profile provides a whole view of the target microorganisms that can potentially contaminate to food products. The aim of this study is therefore focused on the population of microflora and identifying the dominants bacterial genus adhered to equipment surfaces at food processing plant. Adhering microflora was identified by installing the stainless steel coupons on the surface of equipment. Fourteen stainless steel coupons were attached on 7 different locations at food processing plant and then coupons were collected at 1, 2, 3, 5 and 9 weeks. The samples were enumerated the number of bacteria adhesion by swab test and spread on different selective media. More than 600 isolates from various agar plates were identified by Maldi-biotyper machine. The total viable count and Pseudomonas spp. count were found on biofilm, ranging from 103 to 104 cfu/cm2. After 5 weeks the highest count (106 cfu/cm2) was observed on coupons attached in the trolley using for carry the broth. A total of 428 isolates showed a high proportion of Gram negative rods (92%), the predominant genus attached to the surfaces was Raoultella spp. (28 %), Pseudomonas spp. (28%) and Serratia spp. (14%). It is noted that microflora adhered on stainless steel coupons were a multi-species organisms, which include foodborne pathogenic as well as spoilage bacteria. Listeria spp. and Staphylococcus spp. were isolated from the food processing plants when certain conditions are met such as humidity and nutrient. These data support that the appropriate cleaning and sanitizing procedure should be strictly applied to assure high safety in food processing.
